rough protocol of C2C12 transfection:

grow cells in 12-well plate to ~70% confluency in 20% Cos FBS no PSG per well, add 5uL lipofectamine to 50 uL Optimem and 2ug DNA to another 50 uL Optimem, sit 5 minutes separately then combine and sit for 20 minutes change C2C12 into Optimem media, add 100 uL of DNA/lipofectamine, incubate for 6 hours at 37C switch to normal media (differentiation or Cos FBS + PSG depending on cell death).
